Composition Each tablet contains: Tofisopam 50 mg.
Pharmacological Actions
Tofisopam belongs to Homophthalazines (formerly called 2,3-benzodiazepines). It has been shown by in vitro receptor displacement studies that homophthalazines do not compete for dopamine receptors, serotonin receptors or binding sites of diazepam or GABA. The binding site of homophthalazines differ from the 1,4-benzodiazepine receptor binding sites.
Tofisopam, during clinical trials, exhibited an effective anxiolytic action and a characteristic psycho-autonomic stabilizing effect. It is therefore, can be considered as a “Harmonizing Tranquillizer”.
It has no sedative, muscle relaxant or an anticonvulsant effects. It does not impair, in fact it may even improve, the psychomotor and intellectual functions. No abuse or dependence develops during its usage. Therefore, it is safely discontinued without fear of withdrawal effects whatsoever.
Contrary to the cardiodepressive effect of other benzodiazepine derivatives, Tofisopam has an important coronary dilator effect. Besides its use as a tranquillizer, it may be of therapeutic value as anti-angina adjuvant as well.
Due to the lack of any central muscle relaxant action, No deprine can be successfully used in patients with myasthenia gravis, myopathy, neuropathic muscular atrophy.
Pharmacokinetics Tofisopam is well absorbed from the stomach and duodenum and reaches a maximum concentration in the blood in 1 to 1.5 hours following its oral administration. In chronic treatment no signs of accumulation were seen. Tofisopam passes through enterohepatic circulation and its metabolites are mainly excreted in faeces.
Indications
No deprine is indicated to relief autonomic manifestations of anxiety such as: 1- Cardio-vascular manifestations: Tachycardia, palpitations, systolic hypertension and psychogenic cardiac pain (cardiac neurosis). 2- Gastro-intestinal manifestations: Difficulty in swallowing, heart burn, vomiting and diarrhea, abdominal distension, indigestion, constipation and anorexia. 3- Psychological and nervous manifestations: Headache, tremors, hyper-reflexia and hyper-activity, “pains allover” secondary to muscular spasm. 4- Sexual manifestations: Impotence, pre-mature ejaculation and frigidity. 5- Skin manifestations: Alopecia, vitiligo, rashes, urticaria, eczema, neurodermatitis, itching, .. etc. 6- Gynaecological manifestations: Dysmenorrhoea, premenstrual tension and climacteric syndrome. 7- Urinary manifestations: Frequency of micturition and incontinence. 8- Respiratory manifestations: Sighing, dyspnea, tightness in chest, hyperventilation that may lead to respiratory alkalosis resulting sometimes in tetany and numbness in the extremities.
No deprine is indicated as adjuvant in “Psycho-somatic Disorders” such as: Coronary artery disease, cardiac arrhythmias, hypertension, peptic ulcer, ulcerative colitis, IBS, bronchial asthma, …. etc.
Side Effects Rare and include: Temporary gastrointestinal complaints, itching and exanthema. When exanthema appears, treatment should be discontinued.
Usage in Pregnancy Teratogenic effects have not been recorded so far, nevertheless, in the first three months of pregnancy, the use of Tofisopam is not recommended.
Dosage One tablet two to three times daily according to the case and the physician's directions.
Package Boxes, each contains two strips. Each strip contains ten tablets.
